The Department of Bioinformatics at the University Medical Center of Göttingen, has been established by November 2002. The Research focus concerns three major areas: (1) Theoretical analyses of regulatory networks in eukaryotic cells, (2) development of practically applicable routines in the areas of gene regulation and signal transduction, in particular for the interpretation of microarray data, and (3) providing integrated Web-based bioinformatics services. Important activities in the first area are modeling of genetic and signalling networks, description of the signals involved, and characterisation and identification of genomic sequence elements and their combinatorics. The second area, which is the most relevant for the project proposed here, comprises development of databases on disease-relevant aberrations in signal transduction components, of a system describing intercellular communication pathways, and of an integrated view of intracellular (signalling, genetic and metabolic) and intercellular (e.g., endocrine) networks. The third area includes the development of an expression data analysis pipeline. Amongst the most important achievements of the group in the past are a series of databases (TRANSFAC, COMPEL, PathoDB, S/MARt DB, TRANSPATH, CYTOMER, PathoSign) and a number of software tools. EndoNet is our new database on endocrine cell-to-cell communication. It enables the analysis of human intercellular regulatory pathways and aims at bridging multiple intracellular and intercellular regulatory networks. Contribution to the proposed project: 1) Modeling of lipid-related intra- and intercellular pathways and networks, with a special emphasis on perturbations caused by diseases. 2) Acquisition and mining of data about lipid-derived components and processes in regard to different organs/tissues (intercellular relationships). 3) Organizing and storing these data within a public database (data curation). That will be done within the framework of our EndoNet database on endocrine regulation, as well as our CYTOMER ontology of cells, structures, and organs of the human organism. 4) Creating and analyzing an integral network of lipid-related communications between different organs/tissues within the whole organism. 5) Contributing to knowledge and hypothesis
