TaqMan Low-Density Array (TLDA)
TaqMan Expression analysis is based on common 384 well plates and assays on demand, it allows us to monitor mRNA expression with 25ng of total RNA per well. Predesigned assays are a collection of over 600000 probe and primer sets for quantitative gene expression analysis of human, mouse, rat, Arabidopsis and Drosophila genes. It is also possible to design custom TaqMan gene expression assays for any gene and any genome.
One major approach will be the use of TaqMan Low-Density Arrays (TLDA) for the above described lipid pathways. The TLDA is based on Applied Biosystems 7900HT Micro Fluidic Card. This method allows simultaneous analysis of 47 or 94 human genes and the reference genes 18S rRNA in 2 different samples per run. with only 2ng of total RNA per well. Thus the 2-μl-small-volume design of each reaction well substantially decreases sample and reagent consumption, and the precaptured primers and probes save time by reducing labor-intensive This has already been set up for all known ABC-transporters (Langmann, Mauerer and Schmitz, Clin Chem 2006) and the Fatty Acid Metabolism. This will allow a standardized comparison of whole pathways in different patients groups.
